PRODUCTS FOR MOLECULAR BIOLOGY

 

 

IoI offers a comprehensive range of radiolabeled nucleotides and amino acids for life science research. With over 30 years experience in the radiolabeling of biomolecules, high quality product range has been developed. The pack sizes, formulation, and specification are intended to meet the requirements of the majority of customers.

 

SUPPLYING

32P, 33P and 35S Nucleotides

32P, 33P and 35S radionucleotides are supplied in aqueous solution at 10 mCi/ml (370 MBq/ml). The new formulated SteadyBlueä and SteadyClearä nucleotides are supplied in stabilized (and colored) aqueous solution and have the same radioactive concentration.

35S-Amino Acids

The in vitro translation grade 35S-Methionine is supplied at 10 mCi/ml (370 MBq/ml), 15 mCi/ml (555 MBq/ml) or 50 mCi/ml (2000 MBq/ml) radioactive concentration in 50 mM TRIS-buffer, pH 7.4 (containing 10 mM 2-mercaptoethanol).

Isolabelä is a mixture of L-[35S]Methionine and L-[35S]Cysteine amino acids. The mix is for in vitro protein labeling and supplied in a sterile aqueous solution containing 50 mM L-Lysine and 10 mM 2-mercaptoethanol at 15 mCi/ml (555 MBq/ml). Both 35S-Methionine and IsoLabelä are also available in SteadyBlueä formulation, i.e. stabilized and colored form.

            NucleoTipä products

NucleoTipä is a novel carrier and delivery system in a convenient solution-free format. NucleoTipä product are identical with the formerly available Redivue Tip™ (GE Biosciences) used to be OEM products of IoI. NucleoTipä is a pipette tip that contains reaction-sized amounts of radiolabeled nucleotides in a highly visible yet solution-free form. An anion exchange film, which coats the inner wall of the tip, reversibly immobilizes and protects the labeled nucleotides until ready for use. Reaction-sized aliquots of radionucleotide are pre-loaded into a pipette tip and reversibly immobilized on to the interior wall for elution directly into a reaction mixture. The risk of spillage is minimized and the highly visible dye aids handling. The performance of NucleoTipä nucleotides is equivalent to SteadyBlue products and provides a good method for delivering a set amount of radioactivity to biological assays.

 

PACKAGING

Both the conventionally formulated and the stabilized radionucleotides and 35S-amino acids are shipped refrigerated in polystyrene containers filled with dry ice. The solid carbon dioxide ensures that under normal conditions of temperature and pressure, the contents remain frozen for at least 72 hours. Stabilized products are filled into a vial equipped with DropBlockä insert to minimize the loss caused by the adherence of solution (typically £ 1 µl) during handling.

IsoPackä the new packaging system has been designed to provide optimum convenience and safety. The products are packaged in a color-coded system.

NucleoTipä is available in packs of five tips, pre-loaded with reaction-sized aliquots of 32P-labelled nucleotide and shipped in a specially designed container.

PRODUCTION

Labeled nucleotides are produced in a wide variety of specific activity, radiolabel and formulation to assure the achievement of appropriate sensitivity, resolution and convenience in assays.

32P, 33P and 35S labeled nucleotides are produced by a novel, improved technology from carrier-free processed isotopes by a full-enzymatic method providing biologically active products with high specific activity. 35S -L-Methionine is produced with high specific activity and biologically active form (L isomer of Methionine). IsoLabelä 35S-amino acid mix is produced from a hydrolysate of E. Coli grown in presence of carrier-free 35S-sulfate. To ensure a high standard of radiochemical and chemical purity both nucleotides and amino acids are purified by HPLC.

 

QUALITY CONTROL

QC is an integral part of production. IoI employs manufacturing and quality assurance systems to ensure that our products consistently meet the standards you require. Each batch of the products is analyzed for both purity (HPLC, TLC, column chromatography, etc.) and biological activity (Random Priming, 5’-end labeling, Transcription, and Translation)

 


AVAILABILITY

32P nucleotides are available for shipment weekly from fresh lots. All 32P nucleotides are reference dated for Friday of the week following the production. 33P-nucleotides and 35S-compounds are available biweekly (every odd week) from fresh lots. All 33P nucleotides are reference-dated for Friday of the week following the shipping. All 35S-compounds are reference-dated for Monday 3 weeks following the production.

The products marked with * are shipped according to mutual accommodations.

 

STORAGE AND USAGE RECOMMENDATION

 

The whole nucleotide production range is available in three different way formulated compositions: conventional (Code: FP-, or FF-), as SteadyBlueä, i.e. colored and stabilized (Code: SBP-, or SBF-) or as SteadyClearä, i.e. colorless, stabilized (Code: SCP- or SCF-) form. The 35S-amino acid products are available in standard and SteadyBlueä formulation.

Conventionally formulated radionucleotides and 35S-compounds should be stored under -10 °C and -60 °C, respectively. Aliquoting of these products is recommended to avoid repeated freezing and thawing. SteadyClearä and SteadyBlueä products contain a proprietary stabilizer that means they can be kept in the fridge in liquid form. More than 20 years experience has justified that there are no difference in biological activity between the conventionally formulated and stabilized products. If the visualization is important use the SteadyBlueä series. The conventionally formulated or the SteadyClearä series is recommended if an inert dye can cause any trouble in your application.

Due to the innovative product design NucleoTip does not contain chemical stabilizers and can still be stored at 4 °C without any compromise on performance. NucleoTip products are supplied in packs of five pre-loaded pipette tips. The inner wall of each tip is coated with an anion exchange film, which immobilizes the radioactivity, and is then simply eluted into a reaction as required.
